NAD+ | Nicotinamide Adenine Dinucleotide — Research Grade
NAD+ (Nicotinamide Adenine Dinucleotide) is a dinucleotide coenzyme present in all living cells. It serves as both an electron carrier in metabolic redox reactions and a substrate for NAD+-consuming enzymes including sirtuins (SIRT1-7), PARPs, and CD38. NAD+ levels decline approximately 50% between ages 40-60 in human tissue samples.
Mechanism of Action
- Sirtuin activation — NAD+ is a required co-substrate for SIRT1/SIRT3 deacetylase activity, regulating mitochondrial biogenesis, DNA repair, and inflammation
- PARP-1 substrate — fuels poly-ADP ribose polymerase-mediated single-strand DNA break repair
- CD38 regulation — CD38 is a major NAD+-consuming enzyme upregulated in inflammation and ageing; targeting CD38 restores NAD+ in aged tissue models
- Mitochondrial function — as an electron carrier (NAD+/NADH), it is central to Complex I of the electron transport chain
Research Areas
NAD+ is studied in ageing biology, neurodegeneration, metabolic disease, cardiovascular disease, and DNA repair. Preclinical NMN/NR supplementation studies have driven interest in NAD+ restoration as an ageing intervention. Clinical trials in humans are ongoing.
