MOTS-c is a 16-amino acid peptide encoded within the mitochondrial 12S rRNA gene (MT-RNR1), making it one of the few mitochondria-derived peptides (MDPs) identified in humans. It translocates to the nucleus during metabolic stress and directly modulates gene expression related to glucose homeostasis.

Mechanism of Action

  • AMPK activation — activates AMP-activated protein kinase, the master metabolic switch governing glucose uptake and fatty acid oxidation
  • Folate cycle modulation — inhibits the folate cycle and de novo purine synthesis, triggering AICAR accumulation and downstream AMPK signalling
  • Exercise-mimicking — plasma levels increase with physical exercise; supplementation mimics exercise-like metabolic adaptations in rodent models
  • Insulin sensitisation — improves insulin sensitivity in high-fat diet and diabetic rodent models
