NAD+ (Nicotinamide Adenine Dinucleotide) is a dinucleotide coenzyme present in all living cells. It serves as both an electron carrier in metabolic redox reactions and a substrate for NAD+-consuming enzymes including sirtuins (SIRT1-7), PARPs, and CD38. NAD+ levels decline approximately 50% between ages 40-60 in human tissue samples.

Mechanism of Action

  • Sirtuin activation — NAD+ is a required co-substrate for SIRT1/SIRT3 deacetylase activity, regulating mitochondrial biogenesis, DNA repair, and inflammation
  • PARP-1 substrate — fuels poly-ADP ribose polymerase-mediated single-strand DNA break repair
  • CD38 regulation — CD38 is a major NAD+-consuming enzyme upregulated in inflammation and ageing; targeting CD38 restores NAD+ in aged tissue models
  • Mitochondrial function — as an electron carrier (NAD+/NADH), it is central to Complex I of the electron transport chain
