L-Glutathione (γ-L-glutamyl-L-cysteinyl-glycine; GSH) is the most abundant intracellular antioxidant and the central hub of the cell’s redox regulatory network. With an estimated intracellular concentration of 1–10mM, GSH participates in over 200 enzymatic reactions, serves as the primary substrate for glutathione peroxidases (GPx), and mediates protein S-glutathionylation as a post-translational modification. Mechanism of Action
- ROS scavenging — directly neutralises superoxide (O₂•⁻), hydrogen peroxide (H₂O₂), and lipid peroxides via GPx-mediated reduction
- Glutaredoxin system — maintains protein thiol redox state through reversible S-glutathionylation, regulating kinase activity and transcription factors
- Phase II detoxification — conjugates electrophiles and xenobiotics via glutathione-S-transferase (GST) for urinary/biliary excretion
- Immune modulation — regulates lymphocyte proliferation, dendritic cell function, and cytokine balance through intracellular thiol status
